4-Nitrophenyl β-D-galactopyranoside (PNPG), with the CAS number 3150-24-1, is a colorless to white crystalline powder that is slightly soluble in water and more soluble in methanol, warm ethanol, dimethyl sulfoxide, and dimethylformamide. It is insoluble in ether. This compound is widely used as a substrate to assess the activity of glycosaminoglycan (GAG)-degrading enzymes and to study the kinetic properties of recombinant enzymes such as Leuconostoc mesenteroides glycosidase (BgLm1). When hydrolyzed by β-D-galactosidase, PNPG yields a yellow-colored product, 4-nitrophenol, which can be easily measured at 405 nm, making it a useful tool in enzyme-linked assays.
PNPG is also used in the preparation of substrate solutions for various biochemical analyses.
