Olmesartan is an angiotensin II receptor blocker (ARB) used to treat hypertension (high blood pressure). It works by blocking the action of certain natural substances that tighten the blood vessels, allowing the blood to flow more smoothly and the heart to pump more efficiently. Olmesartan is available under various brand names, including Benicar, Azor, and Tribenzor.
Mechanism of Action: Olmesartan blocks the AT1 receptor, which is responsible for the vasoconstrictive effects of angiotensin II. This leads to vasodilation and a subsequent decrease in blood pressure. It has more than a 12,500-fold greater affinity for the AT1 receptor than for the AT2 receptor and does not affect the response to bradykinin, unlike ACE inhibitors.
Indications: The drug is primarily indicated for the treatment of hypertension, either alone or in combination with other antihypertensive agents. It can also be used off-label for the management of Type 2 Diabetes-associated nephropathy, heart failure, and post-myocardial infarction, particularly in patients who are unable to tolerate ACE inhibitors.
Dosage and Administration: The usual starting dose for adults with hypertension is 20 mg once a day, which can be adjusted up to 40 mg based on the patient's response. For pediatric patients, the dose is based on body weight and must be determined by a healthcare provider.
