Griseofulvin is a well-established antifungal medication that has been in use since the late 1950s. It is derived from the Penicillium griseofulvum species of fungi and is primarily used to treat dermatophyte infections, such as ringworm (tinea corporis, tinea pedis, tinea cruris, tinea barbae), athlete's foot, and fungal infections of the scalp, fingernails, or toenails .
Griseofulvin is considered fungistatic, meaning it inhibits the growth of fungi rather than killing them. It is believed to exert its action by binding to fungal microtubule proteins, particularly tubulin, which disrupts the normal process of fungal cell division during mitosis. This leads to the cessation of fungal growth .
Upon oral administration, griseofulvin is absorbed from the gastrointestinal tract and distributed to the keratin precursor cells. It has a high affinity for keratinized tissues, such as the stratum corneum of the skin, hair, and nails. The drug remains in these tissues for an extended period, providing a sustained effect against fungal infections. The half-life of griseofulvin in the blood is approximately 9 to 21 hours, and it is metabolized in the liver and excreted in the urine and feces as metabolites .
