Mechanism of Action: Synthetic vitamin D3 analog that binds to vitamin D receptors (VDR) in keratinocytes and immune cells. It regulates cell differentiation and proliferation while reducing inflammation.
Bioavailability: Minimal systemic absorption when applied topically.
Primary Use: Treatment of plaque psoriasis (psoriasis vulgaris).
Off-label Uses:
Scalp psoriasis
Mild-to-moderate chronic plaque psoriasis in adults and children (in some formulations)
Topical cream/ointment/scalp solution: Apply a thin layer once or twice daily to affected areas.
Maximum weekly dose: 100 g (to avoid hypercalcemia risk).
Combination therapy: Often used with corticosteroids for enhanced efficacy.
Common:
Skin irritation, burning, itching, or dryness at the application site.
Mild erythema (redness) or peeling.
Serious (rare):
Hypercalcemia (elevated calcium levels) with excessive use.
Worsening of psoriasis (paradoxical reaction).
