Omeprazole is a proton pump inhibitor (PPI) used to manage and treat conditions such as heartburn, peptic ulcer disease, gastroesophageal reflux disease (GERD), Zollinger-Ellison syndrome, and erosive esophagitis, among others. It works by blocking the final step of acid production in the stomach's parietal cells, thus reducing the amount of acid made. Omeprazole is available in various forms, including delayed-release capsules, tablets, and oral suspension, and is typically taken once a day, preferably in the morning and before a meal.
The drug is metabolized by the hepatic cytochrome P450 enzyme system, primarily via CYP2C19 and CYP3A4 isozymes, and is excreted mainly in the urine. It has a short half-life but its effects can last longer due to its irreversible inhibition of the H+/K+ ATPase enzyme in the stomach's parietal cells.
Common side effects of omeprazole include headache, abdominal pain, diarrhea, nausea, and vomiting. More serious side effects can include kidney problems, symptoms of a new or worsening lupus, and an increased risk of bone fractures with long-term use. Omeprazole should not be used by patients with a history of hypersensitivity to the drug or any of its excipients.
